RNA interference reveals the escape response mechanism of <i>Paramecium</i> to mechanical stimulation

چکیده
In Paramecium, a mechanical stimulus applied to the posterior portion of cell causes transient increase in membrane permeability potassium ions, transiently rendering hyperpolarized state. Hyperpolarization Cyclic adenosine monophosphate (cAMP) concentration cilia, resulting fast-forward swimming cell. Schultz and coworkers (1992) reported that unique adenylate cyclase (AC)-coupled channel is involved reaction underlying this response, which known as “escape response.” However, AC responsible for remains be identified. Moreover, molecular linkage between mechanoreception activation has not been elucidated adequately. Currently, we can perform an efficient simple gene-knockdown technique Paramecium using RNA interference (RNAi). one several model organisms whole-genome sequences have elucidated. The RNAi whole genome derived from database (ParameciumDB) investigate types proteins elicit specific biological responses compare them with those other organisms. review, describe applications elucidating mechanism escape response identifying reaction. findings study highlight advantages ParameciumDB.

Escape behavior in response to mechanical stimulation of hindwing in cricket, Gryllus bimaculatus.
We studied behavioral responses of the cricket, Gryllus bimaculatus, to mechanical stimulation of the hindwing tip using three different kinds of stimuli: touching, bending and pinching. The most characteristic was a sequence of initial jump-like movements and subsequent running steps, that is referred to as escape behavior in this study. Touching stimulus elicited the escape behavior in 52% of...
متن کاملEscape response of planktonic protists to fluid mechanical signals
The escape response to fluid mechanical signals was examined in 6 protists, 4 ciliates and 2 dinoflagellates. When exposed to a siphon flow, 3 species of ciliates, Balanion comatum, Strobilidium sp., and Mesodinium pulex, responded with escape jumps. The threshold deformation rates required to elicit an escape ranged between 1.8 and 3 s–1. Escape speeds varied between 100 to 150 body length s–1...
متن کاملParallel RNA interference screens identify EGFR activation as an escape mechanism in FGFR3-mutant cancer.
UNLABELLED Activation of fibroblast growth factor receptors (FGFR) is a common oncogenic event. Little is known about the determinants of sensitivity to FGFR inhibition and how these may vary between different oncogenic FGFRs. Using parallel RNA interference (RNAi) genetic screens, we show that the EGF receptor (EGFR) limits sensitivity to FGFR inhibition in FGFR3-mutant and -translocated cell ...
متن کاملEnhancement of RNA Interference Effect in P19 EC Cells by an RNA-dependent RNA Polymerase
Background: RNA interference (RNAi) is a phenomenon uses double-stranded RNA (dsRNA) to specifically inhibit gene expression. The non-specific silencing caused by interferon response to dsRNA in mammalian cells limits the potential of utilizing RNAi to study gene function. Duplexes of 21-nucleotide short interfering dsRNA (siRNA) inhibit gene expression by RNAi. In some organisms, siRNA can als...
متن کاملan introduction to rna interference (rnai)
the discovery of rna interference (rnai) in recent years has revolutionized biological and medical research. rnai is the sequence-specific gene silencing triggered by double-stranded rna (dsrna). until a couple of years ago only biologists working with caenorhabditis elegans and plants could use rnai technology to gain insight into gene function……
